An epilepsy‐associated ACTL6B variant captures neuronal hyperexcitability in a human induced pluripotent stem cell model
The authors investigated the pathophysiological mechanism underlying the pathogenic variants in ACTL6B patients. Using patient-derived neurons they were able to show reduced protein stability, and dysregulation of target gene expression important for neuronal disease and development. MEA recordings showed elevated electrophysiological activity in the patient-derived neurons consistent with the patient phenotype.

Electrodes can be disabled using the right-click menu in the Continuous Waveform Plots, Spike Plots, or Cardiac Beat Plots panels in AxIS Navigator. To disable or enable an electrode in the selected well, right click on an electrode plot and select Disable Electrode to turn it off or on. A blank plot indicates an electrode has been turned off. Alternatively, an electrode can be turned on or off by double-clicking on it in the Plate Map Editor. Wells can be turned on or off with the toggle switch under Well Information. Disabled wells appear black and disabled electrodes appear gray in the Plate Map Editor. Turning off a well or electrode will prevent it from being recorded or analyzed, reducing the file size.
